Commentsmajor h3, h4 germline KO cells were rescued with hy3. Hybrid H3 contruct with H3(1-75) H3.3 (76-90) H3 (91-135), then transformed with Bsr-mtt1-cna1-gfp construct.

CNA1-GFP seq. was used to replace MTT1 coding seq. Bsr1 cas.

Thaw cells without drugs, then grow them in 60-100ug/ml blasticidin, Use 0.5-1 ug/ml cadmium chloride to express and see the green centromere.
